From and to Hamburg there is at least every hour a train towards and from Southern Germany. These trains stop at the most important cities like: Dortmund, Essen, Dusseldorf, Cologne and Frankfurt. Travelling time is about 4 hours between Hamburg and Cologne.
German railways: Die Bahn
The Hauphtbahnhof (Central railwaystation) is situated near the Frankfurt Fair (Messe) Area. It's also within walking distance of the main Skyskraper area if you don't mind walking a bit.
There is a link to the airport by regional railways, but that's difficult (not clearly indicated). Better is to have an intercity train to Cologne which makes a stop at the airport which has it's own small railwaystation on the main railwayline towards the city of Cologne.
The fastest intercity trains reach Cologne in about 1 hour time. Other intercity trains follow the Rhine river and are a bit slower.
